Aim: To prepare and submit 100 ml of aqueous Iodine Solution (Lugol’s Solution).
Requirements: Iodine, Alcohol, Potassium Iodide, Beaker, Glass rod, Measuring cylinder, etc.
Theory: Lugol’s iodine, also known as aqueous iodine and strong iodine solution, is a medication and disinfectant used for several purposes. Iodine is practically insoluble in water as its solubility is 1: 2150. But in the presence of Potassium Iodide, it becomes soluble due to the formation of a new compound known as poly iodide and acts as a stronger germicidal. Lugol’s iodine, by contrast, has no alcohol and has twice the mass of potassium iodide as elemental iodine. Lugol’s iodine solution is often used as an antiseptic and disinfectant.
Formula:
Ingredients | Standard formula |
Iodine | 50 g |
Potassium Iodide | 100 g |
Purified water | 1000 ml |
Procedure:
- Take Potassium Iodide as required in the beaker.
- Add 5-7 ml of water and stir well.
- Add the required amount of iodine to it and stir well.
- Make up the volume up to 100 ml with alcohol.

Storage: Store in a well-closed container and keep in a cool place.
